A Desire is something essential for which your MC has an intense need to obtain. It creates a sense of resolve to sacrifice less important things in exchange for something much better. When your MC has a Desire that is powerful enough to motivate her to embark on a difficult and often perilous journey, you have a story that needs to be told.

We can visualize the concept of Desire in a diagram. Your MC’s location at the beginning of the story is at Point A and her Desire is at Point B. Since she has a powerful Desire to journey to Point B, she must sacrifice her comforts and resources at Point A.
